If you really want a comma in there, write: The city's population, over the last five years, grew 20%.(making "over the last five years" into a parenthetical expression) 4 is okay; a comma after 2003 would make it more readable but it's not wrong--a lot of people argue over this type of comma.

If I was editing this sentence, I'd put the comma in. 5 is correct--it is the same type of sentence as #4 and some might argue to take out the comma 6 is correct--the semicolon joins two complete sentences Some might argue that the percentages should be spelled out (twenty percent, not 20%), as well as the money (ten million dollars) but you only asked about punctuation.
