Gh Quality Material is the Key For me personally I don't notice this difference unless I have 1080P content. Currently the only options for that are Blu-Ray, HD-DVD, or hooking up your PC. For the movies you have to have a large enough tv relative to the distance away otherwise the human eye is not sensitive enough.

Yes Ive got a 720P tv and my brother has a 1080p tv. Both HD movies and Xbox 360 games look a bit better than it does on my tv - enough better that I wish I had purchased the 1080p TV now... Live and learn I guess. But I can tell you that I can see a noticable difference in clarity on his compared to mine.

Above 42" yes, the difference is quite visible.. 42" is so-so, but 1080i still better As the TV gets larger, the difference with 1080i becomes quite noticeable, assuming you have a real HD picture source coming in.
