I want to transfer ALL data and programs from a PC and a laptop to an external 500GB harddrive. Nice try, but you can't do that completely You will still need for Windows to be installed on your boot disk - Window's can't boot from an external (i.e. USB) drive. Also, many programs place DLLs and similar support files on the \WINDOWS directory of the boot drive, so it can find them.
Furthermore (if you are using XP or Vista) many application related configuration files are loaded either in your Documents and Settings\Local\Applications or \Program Data directories. So you can get around 95% of the stuff off of your main hard drive, but you can never get 100% of it off.

Connect the external hard drive to one of the computers, preferably not the laptop for obvious reasons and then network the laptop to the other computer using a fast ethernet rj45 crossover cable, make sure that you purchase a cross-over cable and not a straight-through. Networking using a rj45 crossover cable. Have fun!.

2 Yeah - I agree with Annlee. I have an external drive that I use as a back up but I've also used it to run certain programs off of for both my desktop and work laptop (just to check that I could). However, if you have all your stuff on your external drive and it fails, then you've just lost everything.
If you do use it as your main drive, make sure you back up your important files to either your laptop or your desktop (or both) - and if they're really important (music, photos, etc. ) also consider burning them to a DVD/CD and storing them in a fire box (which I do) or using a remote storage service (several available online but it may cost you a monthly fee).
