Own a Nikon D80. Want telephoto lens. Should I choose new Nikon 70-300 VR or Tamron 18-250?

Nikon Lenses I'd stay away from off brand lenses if it's economically feasible. The Nikon 70-300VRII is a nice lens and not outrageously expensive for what you get. It's on my list for next purchase.
Just be careful when you buy as there are two 70-300s on the market. The older "beer can" lens is a lot cheaper but doesn't have VR. It's a very nice lens for the price if you can live without the VR.It was my first (and, so far, only) 300mm telephoto but it will be replaced with the 70-300VRII just as soon as I win the lottery.
If you can afford it, and don't need the 300mm telephoto, look at the 18-200 VRII. It's an amazing lens (as well as twice the price of the 70-300). Since I put that on my D80 I almost never take it off except to use the 50mm prime inside (or if I'm going someplace where it might get busted).
I also have the 55-200VR which is a very nice lens for a very reasonable price. It's much lighter but not quite as versatile as the 18-200VRII. I took that to Portland, ME this summer and got nice shots from closeups of flowers to long shots across the harbor and everything came out very sharp.

With about $500, Nikon 70-300 would be a good choice. If you don't care about VR, you can save even more. Given that you already have a all-around (xx-135) lens, between Tamron 18-250 and Nikon 70-300 VR, I'd choose Nikon 70-300 VR any time.
The Tamron 18 - 250 simply has too much overlap with what you have. A friend of mine uses 70-300 all the time (sometimes he'd carry a second camera for the wide-to-prime range shots), and I've seen a fair amount of great eagle pictures from him. I've read that the optics of Tamron 18-250 is quite impressive, but I really don't know enough to say for sure.
Bare in mind, if you're tempted to get the 18-250 as a replacement of (xx-135), somewhat being a super all-around lens, the lack of VR could hurt you. If you intend to use the lens primarily on a tripod, and really don't care about the VR capability, another option would be hunting for the older Nikon 70-300mm (without VR). They're much cheaper (around $200 I think) and arguably just as good.

Both will have too much image loss for anything larger than 4x6. A rule of thumb is to never buy a zoom with more than 3x focal increase. So, if the lower-end is 70mm, the high end should be 210mm or less.
If the low end is 24mm, the high end should be 72mm or less. Look at Nikon’s (and similar range in Canon) high-grade "glass". 24-70 f/2.8 70-200 f/2.8 120-300 f/2.8 (Sigma) 200-400 f/4 See the pattern?
Super-zooms (4x or more) are sacrificing image quality for focal range. You may get great shots from them on occasion, but that won’t be the norm. There may be an extremely sharp copy of the lens out there, but you’ll have to look for it.
Since you don’t say what lens you do have, I’ll assume something along the 18-55 range (standard starter "kit" lens on many consumer-grade dSLRs). I'm going to recommend a 70-200/210 lens for you, possibly used to save some bucks. If you can stretch it, get the 70-200 f/2.8 with or without VR (I shoot Canon, so I may have the lens wrong).

Go for the Nikon The Nikon has a few advantages over the Tamron. First and most important for a long lens is the VRII feature on the Nikon, which is a huge plus on removing handshake at 300mm. Their marketing material this system can enable you to get a shot that looks about 4 shutter stops faster and I have read reviews on Nikon VR that their claim seem to live up to the real world.
The other feature the Nikon has is an internal focus motor which provides a faster, quieter, and more power efficient autofocus than a pin driven lens like the Tamron. Also not sure if 200 mm would work for you, but Nikon also offers an 18-200mm VRII lens that may be worth looking into. This would give you a very versatile lens for almost all situations in one package.
