Well to make a design like the squarespace, you have to learn how to draw on photoshop. If you want to draw in vector mode, you would have to know Adobe illustrator also. I recommend you to learn photoshop first and when you learn the basic interface of it, you should learn illustrator after that.

If you want to make your site more functional and dynamic, you should also learn adobe flash. Although learning all these can be taught by the tutorials available from youtube for free, you might want to buy a book that teaches you how to web design.
