I am a doctor (15 years in practice) and have given Vitamin B12 to many people - some I think is placebo effect, but some seem to benefit. My job is to be aware of the benefits or side effects of anything I give to any of my patients, and to my knowledge there is no negative to getting too much vitamin B12. It is impossible to be allergic to B12 as it is an essential substance for life (hence a "vitamin" - from the word "vital").

There is no conflict with the amitryptilline either. I will say that the problem with B12 is absorption. It is usually given as an injection because it requires something called "intrinsic factor" to absorb it in the stomach.

People with true B12 deficiency (something called "Pernicious Anemia") can also have specific problems with their stomach (a type of gastritis that can be serious). If you are truly B12 deficient, you should discuss this with your doctor.

I'm so sorry this is happening to you :( Vitamin B supplements are great for when you are feeling a bit tired, assuming it's not contraindicated (such as an allergy), but if you are having possible side effects, it's not a bad idea to try a lower dose (split the tabs, for example, or take a daily multivitamin with a lower amount of vitamin B, and some other vitamins in there as a bonus).
